To take advantage of AGR, I simply can go to Home Depot and show my AGR card when checking out????

No. You must order online to earn points. Some stores (such as Circuit City, Office Depot and Sears) offer you the option of either in-store pickup or delivery, but other stores (such as Staples and Office Max) require the items be delivered to earn points. I'm not sure if HD offers in-store pickup - but I don't believe so.

If I were to purchase a $250.00 Florida pass, would that entitle me to 500 pts.

If you mean 500 AGR points - I believe so, since you get 2 AGR points/$ spent for Amtrak travel, and that would be Amtrak travel!
